Procaine is the oldest aminoester local anesthetic in clinical use. It is a derivative of PABA, with a relatively high pKa and low lipid solubility, which gives it a slow onset and a short duration of action. This reduces its usefulness when compared with other short-acting local anesthetics, such as lidocaine.
